到奇妙的酒店消滅鬼怪…
被邀請到豪華酒店的路易吉一行人。到達的酒店竟然是由「害羞幽靈王」的超級粉絲老闆開的一個陷阱！
就當路易吉在酒店打瞌睡的時候，瑪利歐、碧姬公主、奇諾比奧們被抓走，更被困在畫裡。
為了拯救同伴，有點膽小的路易吉拿起「哎唷‧喂博士」所發明的新型「鬼怪吸塵器」，在奇妙的樓層「鬼怪酒店」展開新的冒險。
